Highly thermostable and alkaline α-amylase from a halotolerant-alkaliphilic Bacillus sp. AB68 BJM
Aygan,Ashabil; Arikan,Burhan; Korkmaz,Hatice; Dinçer,Sadik; Çolak,Ömer.
An alkaliphilic and highly thermostable α-amylase producing Bacillus sp. was isolated from Van soda lake. Enzyme synthesis occurred at temperatures between 25ºC and 40ºC. Analysis of the enzyme by SDS-PAGE revealed a single band which was estimated to be 66 kDa. The enzyme was active in a broad temperature range, between 20ºC and 90ºC, with an optimum at 50ºC; and maximum activity was at pH 10.5. The enzyme was almost completely stable up to 80ºC with a remaining activity over 90% after 30 min pre-incubation. Thermostability was not increased in the presence of Ca2+. An average of 75% and 60ºC of remaining activity was observed when the enzyme was incubated between pH 5 and 9 for 1 h and for 2 h, respectively. The activity of the enzyme was inhibited by...

How can nutritional strategies and feed technologies in pig production affect the logistical costs of manure distribution? R. Bras. Zootec.
Afonso,Esther Ramalho; Nacimento,Rafael Araujo; Palhares,Julio Cesar Pascale; Gameiro,Augusto Hauber.
ABSTRACT The objective with this study was to evaluate the impact of different swine nutritional strategies and technologies, as well as the land spread system, on logistical costs of pig manure distribution. For this, pigs were fed a conventional diet (C0) or a diet supplemented with 0.01% phytase (C1); organic minerals (40% substitution; C2); synthetic amino acids at reduced dietary crude protein levels (C3); or a combination of the three strategies (C4). All pig manure was weighed, then its chemical composition was determined. The mineral values and volume of manure produced were extrapolated to corresponding pig farm sizes in Sao Paulo state (300, 650, and 1000 sows) from real pig manure levels to produce simulations. To determine the logistical cost...

Impacts of three different microdiets on Florida Pompano, Trachinotus carolinus, weaning success, growth, fatty acid incorporation and enzyme activity. ArchiMer
Hauville, Marion; Zambonino, Jose Luiz; Bell, G; Migaud, Herve; Main, Kevan L..
In this study, three microdiets were tested on weaning of Florida pompano larvae: Otohime, Gemma and a reference diet LR803. The experimental system was stocked with 11-day-old larvae, which were co-fed micro-diets and live food from 11 dph to 17 dph then micro-diets only until 28 dph. Survival from 11 dph to 28 dph was similar for all treatments, with an average of 33%. At the end of the trial, the Gemma larvae were significantly longer and heavier than larvae fed the other diets. Significant differences were observed in fatty acid composition of the diets and larvae between treatments. The Gemma larvae incorporated the lowest amount of eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA) and arachidonic acid (ARA). However, they had the highest...

Inactivation of yeast inorganic pyrophosphatase by organic solvents Anais da ABC (AABC)
Grazinoli-Garrido,Rodrigo; Sola-Penna,Mauro.
A number of application for enzymes in organic solvents have been developed in chemical processing, food related conversions and analyses. The only unsolved problem related to nonaqueous enzymology is the notion that enzymes in organic solvent are mostly far less active than in water. Therefore, studies concerning the mechanisms by which enzymes are inactivated by organic solvents would reveal a clear understanding of the structure-function relationship of this phenomenon. Here we analyzed the effects of a series of alcohols (methanol, ethanol, 1-propanol and 2-propanol) and acetone on the activity of yeast inorganic pyrophosphatase. We observed that solvents inactivated the enzyme in a dose-dependent manner. This inactivation is also dependent on the...

Increasing levels of phytase in diets formulated with reduced available phosphorus content supplied to male and female broilers Ciência Rural
Naves,Luciana de Paula; Rodrigues,Paulo Borges; Teixeira,Levy do Vale; Bertechini,Antônio Gilberto; Alvarenga,Renata Ribeiro; Bernardino,Verônica Maria Pereira; Santos,Luziane Moreira dos; Natividade,Yuri Pereira Efrem.
The reduction of the available phosphorus (avP) content in the broiler diet must be followed by the supplementation of adequate phytase. One experiment was conducted with broilers from 15 to 28 days of age to determine the level of necessary phytase when the mashed diet is formulated with 0.213% of avP to enable the results of performance, tibia ash content, and calcium (Ca) utilization similar to those determined for birds fed with diet formulated to meet their nutritional requirements. Thus, 120 broilers were distributed in (4+1) x2 factorial arrangement corresponding to four deficient diets in avP (0.213%) supplemented with phytase (0; 750; 1,500 or 2,250FTU kg-1) plus one positive control diet without phytase (0.426% of avP), supplied to male and...

Low carbon/nitrogen ratio increases laccase production from basidiomycetes in solid substrate cultivation Scientia Agricola
D'Agostini,Érica Clarissa; Mantovani,Talita Rafaele D'Agostini; Valle,Juliana Silveira do; Paccola-Meirelles,Luzia Doretto; Colauto,Nelson Barros; Linde,Giani Andrea.
Basidiomycetes are laccase producers used for hydrolysis of lignocellulosic byproducts in fermentative processes and could be used on biofuel production or ruminant feeding. The objective of this study was to evaluate the effect of concentrations of non-protein nitrogen sources on laccase production and mycelial growth of Pleurotus ostreatus, Lentinula edodes and Agaricus blazei. The fungi were grown on soybean hulls to which urea (U), ammonium sulfate (AS) or mixture of AS:U (1:1) were added to achieve carbon/nitrogen (C/N) ratios of 5, 15, 20 or 30. The average longitudinal mycelial growth was measured and laccase activity was determined by the oxidation of 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid. Higher C/N ratios increased mycelial growth...

Nitrate reductase activity in leaves and stems of tanner grass (Brachiaria radicans Napper) Scientia Agricola
Cazetta,Jairo Osvaldo; Villela,Luciana Cristine Vasques.
Tanner grass (Brachiaria radicans Napper) is a forage plant that is adapted to well-drained soils or wetlands, and responds well to nitrogen (N) fertilization. The assimilation of N involves the nitrate reductase (NR) enzyme, and its activity seems to be dependent on N supply. Molybdenum (Mo) is also important because it is a cofactor of NR. In this study, the variables of an in vivo assay were optimized for measuring nitrate reductase activity (NRA) in the leaves and stem tissues. This method was used to evaluate NO3- metabolism in plants fertilized with NaNO3, NH4Cl or urea, in association with or without application of H2MoO4, aiming to provide guidelines for N management of this species. The best conditions to determine NRA involved the incubation of...
